1.A Hormone Fragment That Lost Its Hormone
The origin of Semax is a masterclass in rational peptide design. It was engineered from the 4-10 fragment lineage of Adrenocorticotropic Hormone (ACTH), specifically targeting the ACTH(4-7) core (Met-Glu-His-Phe). While the parent hormone is famous for triggering the body's systemic stress-hormone cascade, researchers discovered that this specific core carried the hormone's neurotrophic and cognitive benefits without any of its hormonal activity.
The true breakthrough was structural. The parent fragment is rapidly cleaved by enzymes in the body. To solve this, scientists at the Institute of Molecular Genetics of the Russian Academy of Sciences added a Pro-Gly-Pro (PGP) tripeptide to the C-terminal. This addition creates steric hindrance against peptidases, significantly extending the peptide's half-life and allowing it to remain active for 20–24 hours in biological models.
